Objective@#The purpose of this study was to investigate the differences in the anti cyclic fatigue performance of Woride KS (WKS), Proteper Gold (PTG), and Hyflex CM (HCM) nickel titanium instruments with different tip diameters in curved root canal models, and to provide reference for the targeted selection of suitable nickel titanium instruments in clinical preparation of curved root canals.

El sistema dispone de un set básico de 4 limas: Z1, Z2, Z3 y Z4, con un D0 de 0,14 mm, 0,17 mm, 0,19 mm y 0,25 mm respectivamente y conicidad variable que oscila entre el 2 y 10 %. ABSTRACT

NiTi endodontic instruments for glide path are the most susceptible to fractures inside the root canal, mainly as a result of high torsional stress. Objective: The present study investigated the resistance to torsion and angular deflection of instruments destined for the glide path: ProGlider #16.02; T-File #17.02 and the MK Life #16.02. Materials and methods: Thirty rotating NiTi glide path instruments (n=10) with 25mm lengths were selected. The torsion test was performed based on ISO 3630-01 (1992). Three millimeters from the tip of the instruments, it was attached to a small load cell by a lever arm connected to the torsion shaft. Torsional strength and angular deflection were evaluated. Fracture surfaces were examined by scanning electron microscopy with magnifications of 1000x and 5000x in the cross section, and 50x in the lateral section. Statistical analysis was performed using the Kruskal-Wallis H test, followed by the Down's post hoc test. Results: The ProGlider instrument showed greater torsional strength (p<0.05) compared to the T-File (p<0.05) and MK Life (p<0.05), respectively. However, the T-File showed greater angular deflection (p<0.05) than the other groups tested. Conclusion: It can be concluded that the ProGlider instrument presented greater torque for the fracture, while the T-File instrument presented greater angular deflection.

Resumen El objetivo del estudio fue comparar la extracción de ADN de quistes de Acanthamoeba sp. con un método disponible comercialmente y cuatro no comerciales utilizando tratamiento térmico y ultrasonido para la amplificación por una reacción en cadena de la polimerasa (PCR) convencional, reduciendo tiempos de preparación y extracción de las muestras, como una herramienta para el diagnóstico en el laboratorio clínico. Se utilizó una cepa de Acanthamoeba, genotipo T4, cultivada en agar no nutritivo. Los quistes para analizar, en tres períodos de enquistamiento, se almacenaron a temperatura ambiente. Se extrajo ADN mediante cinco métodos: pretratamiento térmico, ultrasonido y combinaciones de ellos. La PCR se llevó a cabo utilizando cebadores específicos JDP1/JDP2. La concentración y pureza del ADN extraído con los protocolos evaluados revelaron diferencias estadísticamente significativas (p<0,0001). El método E (comercial), el A (térmico) y el B (ultrasonido) lograron los mejores rendimientos en la amplificación del fragmento específico de Acanthamoeba sp. por la PCR convencional.


Abstract The objective of the study was to compare the DNA extraction of Acanthamoeba sp. cysts with a commercially available method and four non-commercial ones, with heat and ultrasound treatment that allows amplification by conventional polymerase chain reaction (PCR), reducing sample preparation and extraction times, such as a tool for diagnosis in the clinical laboratory. To this aim, a strain of Acanthamoeba T4 grown on non-nutrient agar was used. Plates with cysts at three different encystation times were stored at room temperature until the study was carried out. DNA was extracted with five methods that included pretreatments (thermal and ultrasound) or combinations of them. PCR was performed using specific primers JDP1/JDP2. Concentration and purity of DNA revealed statistically significant differences (p<0.0001) between methods. Method E (commercial), method A (thermal) and B (ultrasound) got the best yields in amplifying the specific fragment of Acanthamoeba sp. by conventional PCR.

Os cistos para analisar foram armazenados em temperatura ambiente, correspondendo a três períodos de encistamento. O DNA foi extraído por cinco métodos: pré-tratamento térmico, ultrassom e combinações deles. A PCR foi realizada usando iniciadores específicos JDP1/JDP2. A concentração e a pureza do DNA extraído com os protocolos avaliados revelaram diferenças estatisticamente significativas (p<0,0001). Os métodos E (comercial), A (térmico) e B (ultrassom) alcançaram os melhores rendimentos na amplificação do fragmento específico de Acanthamoeba sp. por PCR convencional.

ABSTRACT

Objective@# To evaluate the effect of heat treatment on the bonding strength of pure titanium formed by selective laser melting (SLM) and porcelain.@*Methods@#Ninety-six pure titanium specimens were laser machined to meet ISO 9693 standards. The specimens were divided into a heat treated group (A) and a nonheat treated group (B). According to the porcelain type, the specimens in groups A and B were divided into Super Ti22 (a), Titankeramik (b), and Triceram (c) groups. Then, according to sandblasting pressures of 0.25 MPa (1) and 0.45 MPa (2), they were further divided into Aa1, Aa2, Ab1, Ab2, Ac1, Ac2, Ba1, Ba2, Bb1, Bb2, Bc1, and Bc2 groups. The surface morphology and roughness of the sandblasted specimens were assessed using a laser scanning confocal microscope. After the porcelain was fused, the three-point bending titanium-porcelain bonding strength was tested. A stereomicroscope was used to characterize the titanium-porcelain interfaces and determine the mode of failure.@* Results@# The Vickers hardness of group A specimens (188.21 ± 11.94) was significantly lower than that of group B specimens (204.48 ± 6.32) HV (P<0.05). The roughness value in group A1 (2.90 ± 0.32) μm was significantly lower than that in group A2 (3.43 ± 0.43) μm (P<0.05). Specimens in group B1 (2.62 ± 0.08) μm were significantly smaller than those in group B2 (3.01 ± 0.06) μm (P<0.05). The bonding strength in group Aa1 was (33.75 ± 2.31) MPa, group Aa2 was (36.32 ± 1.44) MPa, group Ab1 was (39.82 ± 2.28) MPa, group Ab2 was (33.74 ± 1.53) MPa and group Ac2 was (38.63 ± 1.36) MPa, which was significantly higher than that in the corresponding groups Ba1 (29.65 ± 1.10) MPa, Ba2 (27.17 ± 2.24) MPa, Bb1 (27.29 ± 1.61) MPa, Bb2 (23.85 ± 0.97) MPa, and Bc2 (35.75 ± 1.93) MPa (P<0.05). With increasing sandblasting pressure, the bonding strength of the titanium ceramic in group Aa2 was significantly higher than in group Aa1, while that in group Ab2 was significantly lower than that in group Ab1 (P<0.05). In groups A, Bc1 and Bc2, the fracture model showed mixed failure, while in groups Ba1, Ba2, Bb1, and Bb2, the model showed interfacial failure.@* Conclusion @# The Vickers hardness of SLM titanium can be significantly reduced by heat treatment. SLM pure titanium after heat treatment is beneficial to combination of the three porcelain types and titanium. The titanium-porcelain bonding strength may be affected by sandblasting pressure.

Objective@#To compare the efficiency and biocompatibility of four different silanes on immobilizing c(RGDfK) peptide on titanium surface.@*Methods @# After alkali-heat treatment (group OH), the titanium surface was treated with 3-aminopropyltriethoxysilane (APTES) (group OHAP), 3-chloropropyltriethoxysilane (CPTES) (group OHCP) (3-mercaptopropyltrimethoxysilane (MPTS) (group OHMPT) and 3-isobutyryloxy propyltrimethoxysilane(γ- MPS) (group OHMPS) to immobilize the c(RGDfK) cyclic peptide and constructa titanium-silane-c(RGDfK) coating. The NT group was the blank control group. The surface morphology and wettability of the coatings were detected using scanning electron microscopy and contact angle measurement. The elemental composition of the titanium surface was analyzed using X-ray photoelectron spectroscopy. After fluorescent staining with 4’,6-diamino-2-phenylindole (DAPI) and phalloidin, the adhesion of mouse preosteoblast MC3T3-E1 cells on the surface of the materials was observed using laser confocal microscopy. Cell counting kit-8 (CCK-8) and alkaline phosphatase (ALP) activity assays were used to evaluate the proliferation and osteogenic differentiation of MC3T3-E1 cells on the surface of the materials, respectively. @*Results @#Scanning electron microscope observation showed a spongy-like 3-dimensional network formed on the titanium surface after alkali-heat treatment with silane-c(RGDfK) coating adhesion. The wettability of each group was greatly improved compared to the untreated titanium surface. The element ratios of Si/Ti and amide-N/Ti in the OHMPS group were the highest. The OHAP group exhibited the best cell adhesion effect. The cell proliferation and ALP activity of the OHAP, OHMPT, and OHMPS groups were significantly higher than the control group (P <0.05); there was no statistical difference between the OHCP group and the control group.@*Conclusion @#MPTS, CPTES and γ-MPS covalently immobilized cyclic peptide c(RGDfK) on the titanium surface, which promoted adhesion, proliferation and osteogenic differentiation of MC3T3-E1 cells. Theγ-MPS conjugated C (RGDfK)cyclic peptide exhibited the best effect. MPTS, CPTES and γ-MPS coupled with c(RGDfK) cyclic peptides had similar biological properties.

Introdução: É importante saber se o hipoclorito de sódio (NaOCl) influencia a resistência à fadiga cíclica das limas de níquel-titânio (NiTi). Objetivo: Avaliar a influência de NaOCl 2,5% na resistência à fadiga cíclica de dois sistemas de NiTi. Material e método: 40 instrumentos rotatórios - 20 TruNatomy® (TRU, Dentsply Sirona, Maillefer, Ballaigues, Suíça) e 20 Prodesing Logic2® (PDL2, Bassi, Belo Horizonte, Minas Gerais, Brasil) - foram aleatoriamente distribuídos em 4 grupos experimentais (n = 10) imersos em água destilada (H2O) e NaOCl 2,5% em temperatura a 37°C. Foram submetidos a testes de fadiga cíclica mensurando o número de ciclos para fratura (NCF) e análise dessas superfícies pós-teste em microscópio eletrônico por varredura. Para a análise estatística entre os grupos, foi aplicada a análise de variância (ANOVA), complementada com o pós-teste de Tukey. Resultado: Houve diferença estatística em todos os grupos (P<0.05). Os instrumentos PDL2 obtiveram maior resistência à fratura nas condições em H2O e em NaOCl 2,5% comparados aos instrumentos TRU. Na análise de grupos de instrumentos nas soluções de NaOCl e H2O, foi observado que o NaOCl 2,5% diminuiu o NCF. Conclusão: A resistência à fadiga cíclica dos instrumentos TRU e PDL2 diminuiu com NaOCl 2,5%. Os instrumentos PDL2 foram mais resistentes à fratura em relação aos instrumentos TRU.


Introduction: It is important to know whether sodium hypochlorite (NaOCl) influences the cyclic fatigue resistance of nickel-titanium (NiTi) files. Objective: To evaluate the influence of NaOCl 2.5% on the cyclic fatigue resistance of two NiTi systems. Material and method: Forty rotary instruments - 20 TruNatomy® (TRU, Dentsply Sirona, Maillefer, Ballaigues, Switzerland) and 20 Prodesign Logic2® (PDL2, Bassi, Belo Horizonte, Minas Gerais, Brazil) - were distributed randomly across four experimental groups (n=10) and submerged in distilled water (H2O) or 2.5% NaOCl at 37°C according to allocation. Cyclic fatigue testing was then performed, measuring the number of cycles to fracture (NCF), and post-test surfaces were analysed by scanning electron microscopy (SEM). Statistical Analysis: Analysis of variance (ANOVA) was applied for between-group analysis, followed by Tukey's post-hoc test. Result: A significant difference was observed in all groups (P<0.05). PDL2 instruments showed higher fracture resistance under H2O and 2.5% NaOCl conditions compared to TRU. Analysis of all instrument groups showed that exposure to 2.5% NaOCl decreased the NCF compared to H2O. Conclusion: Cyclic fatigue resistance of the TRU and PDL2 instruments was decreased by exposure to 2.5% NaOCl. PDL2 instruments were more resistant to fracture than TRU instruments.

ABSTRACT

Members of the bZIP transcription factor family play crucial roles in the regulation of plant development, biosynthesis of secondary metabolites, and response to abiotic and biotic stresses. To date, multiple bZIPs have been identified and investigated in numerous plant species. However, few studies have characterized bZIPs from Dimocarpus longan Lour. In this study, nine bZIPs from D. longan were identified from RNA-Seq data and further verified using the NCBI conserved domain search tool and Pfam database. Bioinformatics tools were used to systematically analyse the physicochemical properties, protein structures, multiple sequence alignment, motif compositions, evolutionary relationships, secondary structures, subcellular localization, phosphorylation sites, signal peptides, GO annotations and protein–protein interactions of the DlbZIPs. The expression patterns of the nine DlbZIPs were evaluated by qRT-PCR in roots and leaves and in response to varying durations of a 38C heat treatment. DlbZIP3, DlbZIP5, DlbZIP6 and DlbZIP7 were differentially expressed between root and leaf tissues. All nine DlbZIPs responded to heat treatment in both roots and leaves, but their specific expression levels differed. DlbZIP4 and DlbZIP8 were highly expressed in roots after heat treatment, whereas DlbZIP1 and DlbZIP5 were highly expressed in leaves after heat treatment. These findings lay a foundation for increasing active secondary metabolite content and improving abiotic stress tolerance in D. longan using transgenic technology

Se evaluó el efecto de la temperatura sobre la desnaturalización de proteínas y la reacción de Maillard en leche entera y descremada con lactosa hidrolizada. Las leches hidrolizadas se trataron térmicamente a 100, 110, 120 y 130 °C durante un período de 1 hora y se midió la concentración de glucosa, el grado de pardeamiento y la desnaturalización de proteínas. El grado de dorado en la leche entera varió de 14.4 (100 °C) a 42.6 (130 °C). Para la leche descremada fue de 20.2 (100 °C) a 38.0 (130 °C). La concentración de glucosa en leche entera (47% p/v) y en leche descremada (41% p/v) después del tratamiento térmico (130 °C) mostró una reducción significativa en relación con el control (25 °C). El efecto de la temperatura en la desnaturalización de proteínas en leche entera y descremada en relación con el control (25 °C) fue del 100%. La leche tratada térmicamente con lactosa hidrolizada promovió la desnaturalización de proteínas con un aumento del pardeamiento característico de la reacción de Maillard, lo que afectó la calidad nutricional.


The effect of temperature in protein denaturation and Maillard reaction in whole and skim milk with hydrolyzed lactose was evaluated. Hydrolyzed milk was thermally treated at 100, 110, 120 and 130 °C over a period of 1 hour and glucose concentration, browning degree and protein denaturation were measured. The browning degree in whole milk varied from 14.42 (100 °C) to 42.63 (130 °C) and 20.21 (100 °C) to 38.03 (130 °C) in skim milk. Glucose concentration in whole milk (47% - w/v) and skim milk (41% - w/v) after heat treatment (130 °C) showed a significant reduction in relation to the control (25 °C). The temperature effect in protein denaturation in whole and skim milk in relation to the control (25 °C) was 100%. Thermally treated milk with hydrolyzed lactose promoted protein denaturation with increasing browning characteristic of the Maillard reaction, thus affecting the nutritional quality.

ABSTRACT Objective The aim of this study was to evaluate and compare the effect of heat treatment at different temperatures on the color and flexibility of nickel-titanium endodontic instruments. Methods Thirty-six nickel-titanium K-file endodontic instruments were divided into six groups and submitted to heat treatment at several temperatures between 450ºC and 750ºC. The color was visually observed. Cantilever-bending tests were performed to measure the flexibility of the instruments. The data were statistically evaluated by one-way analysis of variance and Tukey post hoc comparisons. Results It was observed that the color becomes darker as the heating temperature increases. The flexibility is significantly higher after the heat treatment (P < 0.05). Instruments treated at 650ºC and 750ºC had higher flexibility than those treated at lower temperatures (450ºC, 500ºC and 550ºC) (P < 0.05). Conclusion Heat treatment has a significant effect on the surface color and flexibility of nickel-titanium endodontic instruments. The highest flexibility was observed in instruments heated at 650ºC and 750ºC.


RESUMO Objetivo O objetivo deste estudo foi avaliar e comparar o efeito do tratamento térmico realizado em diferentes temperaturas sobre a cor e a flexibilidade dos instrumentos endodà´nticos de níquel-titânio. Métodos Trinta e seis instrumentos endodônticos de níquel-titânio tipo K foram divididos em seis grupos e submetidos ao tratamento térmico com temperaturas variando entre 450ºC e 750ºC. A cor dos instrumentos após tratamento foi visualmente observada. Testes de flexão em 45º foram realizados para medir a flexibilidade dos instrumentos. Os dados foram avaliados estatisticamente através de anà¡lise de vâriancia (ANOVA) com teste post hoc de Tukey. Resultados Observou-se que os instrumentos apresentaram cor mais escura à medida que a temperatura de aquecimento foi aumentada. A flexibilidade é significativamente maior em instrumentos submetidos ao tratamento térmico (P < 0,05). Os instrumentos tratados a 650ºC e 750ºC tiveram maior flexibilidade do que aqueles tratados em baixas temperaturas (450ºC, 500ºC e 550ºC) (P < 0,05). Conclusào O tratamento térmico afeta significativamente a cor apresentada pela superfície dos instrumentos endodà´nticos de níquel-titânio e sua flexibilidade. A maior flexibilidade foi observada em instrumentos tratados nas temperaturas de 650ºC e 750ºC.

As características geométricas e técnicas de fabricação das limas endodônticas afetam o desempenho clínico e sua resistência à fratura, flexibilidade e outras propriedades. Um parâmetro muito explorado na Endodontia é a utilização de tratamentos térmicos, que promovem um aumento na resistência à fadiga e flexibilidade destes materiais. Neste estudo pretendeu-se avaliar o efeito de diferentes processos de tratamento térmico/usinagem nas propriedades físicas, flexibilidade, resistência torcional e à fadiga flexural dos instrumentos Prodesign Logic (PDL), verificando se a sequência de processamento dos instrumentos levaria a comportamentos mecânicos diferentes. Este sistema é fabricado a partir de um método inovador que envolve a realização do tratamento térmico para obtenção de memória de forma no fio NiTi anteriormente à usinagem. A hipótese nula a ser testada é que o momento de aplicação dos procedimentos de tratamento térmico e usinagem não afetariam as propriedades físicas e mecânicas de instrumentos endodônticos de NiTi. Para grupos de comparação, foram utilizados protótipos de instrumentos com as mesmas características geométricas e dimensionais, sendo um grupo de instrumentos sem tratamento térmico aplicado (SEP) e outro com o tratamento térmico realizado de forma convencional, após a usinagem do instrumento (HTP). Foram realizadas caracterização física e microestrutural com ensaios de calorimetria exploratória diferencial (DSC) e testes de difratometria de Raios-X (DRX), além de ensaios mecânicos divididos em testes de torção, dobramento em 45 e resistência à fadiga flexural. Os resultados mostraram que macroscopicamente não houve nenhuma alteração importante nas características dos três grupos de instrumentos, exceto mudanças de cor devido a diferentes sequências de processamento. Os resultados de DSC mostraram que à temperatura ambiente a liga é principalmente austenítica no grupo SEP e que após o tratamento térmico realizado nos grupos PDL e HTP, houve uma mudança nas temperaturas de transformação, com um aumento da temperatura de Af e predominância da liga NiTi em fase R. Os resultados de XDR confirmaram os achados do DSC. As diferenças nos parâmetros medidos entre diferentes tipos de instrumentos nos ensaios de resistência torcional, dobramento a 45º e fadiga flexural foi sempre considerada com um nível de significância de 95%. Os resultados para os ensaios de flexão mostraram que o grupo HTP mostrou-se o mais flexível, com os mais baixos valores de momento de dobramento, seguido por PDL e SEP. Nos resultados dos ensaios de torção foi possível observar que SEP apresentou valores mais altos de torque seguido por PDL e HTP (p<.05). Os ensaios de resistência à fadiga mostraram valores médios de NCF maiores para HTP, seguido por PDL e SEP. Entretanto, um pior comportamento do PDL só seria evidenciado clinicamente se os instrumentos fossem usados um número de vezes maior do que o recomendado pelo fabricante. Pode-se concluir que alterar o processo de fabricação dos instrumentos endodônticos, invertendo-se as etapas de usinagem e tratamento térmico, levou, de fato, a uma mudança no comportamento mecânico de instrumentos Prodesign Logic em relação aos prótótipos.


The geometric characteristics and manufacturing techniques of endodontic instruments affect clinical performance, fracture resistance, flexibility and other properties. A parameter widely explored in Endodontics is the use of heat treatments, which promote an increase in the resistance to fatigue and flexibility of these materials. This study aimed to evaluate the effect of different heat treatment / machining processes on the physical properties, flexibility, torsional strength and flexural fatigue of Prodesign Logic (PDL) instruments, verifying whether the processing sequence of the instruments would lead to different mechanical behaviors. This system is manufactured using an innovative method that involves performing thermal treatment to obtain shape memory in the NiTi wire prior to machining. The null hypothesis to be tested is that the moment of application of the heat treatment and machining procedures would not affect the physical and mechanical properties of NiTi endodontic instruments. For comparison groups, instrument prototypes with the same geometric and dimensional characteristics were used, being one group of instruments without thermal treatment applied (SEP) and another with thermal treatment carried out in a conventional manner, after machining the instrument (HTP). Physical and microstructural characterization were performed with differential scanning calorimetry (DSC) tests and X-ray diffractometry (XRD) tests, in addition to mechanical tests divided into torsion tests, 45° bending and resistance to flexural fatigue. The results showed that macroscopically there were no major change in the characteristics of the three groups of instruments, except for color changes due to different processing sequences. The DSC results showed that at room temperature the alloy is mainly austenitic in the SEP group and that after the heat treatment carried out in the PDL and HTP groups, there was a change in the transformation temperatures, with an increase in the Af temperature and predominance of the NiTi alloy in phase R. The XRD results confirmed the DSC findings. Differences in the parameters measured between different types of instruments in the tests of torsional resistance, bending at 45º and flexural fatigue were always considered with a significance level of 95%. The results for the flexibility tests showed that the HTP group proved to be the most flexible, with the lowest values, followed by PDL and SEP. In the results of the torsion tests it was possible to observe that SEP presented higher values of torque followed by PDL and HTP (p <.05). Fatigue resistance tests showed higher mean NCF values for HTP, followed by PDL and SEP. However, a worse behavior of the PDL would only be evidenced clinically if the instruments were used several times greater than that recommended by the manufacturer. It can be concluded that changing the manufacturing process of endodontic instruments, inverting the machining and heat treatment steps, led, in fact, to a change in the mechanical behavior of Prodesign Logic instruments in relation to the prototypes.

Effect of cooling rate during oxidation treatment on the hardness change by post firing heat treatment of a Pd-Cu-Ga-Zn alloy after simulated porcelain firing was investigated and the following results were obtained. In the firing simulated specimens after quenching during the oxidation treatment, the hardness did not increase by post-firing heat treatment. However, in the firing simulated specimens after slow cooling during the oxidation treatment, post-firing heat treatment for 25 minutes was effective in increasing the hardness (p < 0.05). In the firing simulated specimens after quenching during the oxidation treatment, the significant decrease in hardness during the post-firing heat treatment was attributed to the coarsening of fine precipitates formed in the matrix and plate-like precipitates. In the firing simulated specimens after slow cooling during the oxidation treatment, the increase in hardness during the post-firing heat treatment for 25 minutes was due to precipitation hardening. From these results, it was found that the cooling rate during the oxidation treatment affected the change of hardness by the post-firing heat treatment after the simulated firing. From these results, post-firing heat treatment increased the hardness of the Pd-Cu-Ga-Zn alloy only when the cooling rate during the oxidation treatment was slow.

ABSTRACT

The aim was to evaluate the microbiological, chemical- physical, and shelf-life quality of milk samples after pasteurization (HTST) for 10 days or ultra-high temperature (UHT) treatment for 120 days. Raw milk counts of mesophilic aerobic microorganisms, Staphylococcus spp. and thermotolerant coliforms before HTST and UHT processing were 6.73 and 7.77; 2.84 and 4.30, and 4.68 and 4.37log10, respectively. Pseudomonas spp. were found in raw milk samples. No presence of any other microorganisms studied was detected and no microbial inhibitor was found. Processed samples met microbiological legal requirements. However, aerobic mesophilic counts for HTST pasteurized milk samples stored for 5 and 10 days increased to values comparable to those in raw milk. Composition chemical- physical of all samples were within legal limits. These results demonstrate that, although HTST and UHT processed milk comply with the microbiological standards required by Brazilian law, high microbial counts in raw milk are an issue, possibly due to failures in the early stages of the production chain. Increase in casein macropeptide (CMP), probably because of proteases psychrotrophic bacteria. It is concluded that the quality of raw milk directly influences the progressive increase of the CMP values.(AU)


O objetivo da presente pesquisa foi avaliar a qualidade microbiológica, fisco-química e a vida de prateleira de amostras de leite, após o processo de pasteurização rápida (HTST) ou de ultra-alta temperatura (UHT) durante 10 dias, ou de ultra-alta temperatura (UHT) por 120 dias. As contagens de micro-organismos aeróbios mesófilos, Staphylococcus spp. e de coliformes termotolerantes do leite cru utilizado para tratamentos HTST e UHT foram, respectivamente (log10): 6,73 e 7,77; 2,84 e 4,30 e 4,68 e 4,37. Foi constatada a presença de Pseudomonas spp. no leite cru. Não foi detectada a presença de nenhum outro micro-organismo estudado, e as amostras estavam isentas de inibidores microbianos. Após a pasteurização, todas as amostras apresentaram contagens microbianas compatíveis com os limites legais. No entanto, as amostras de leite pasteurizado apresentaram contagens de aeróbios mesófilos semelhantes ao leite cru após cinco e 10 dias de armazenamento. A composição físico-química de todas as amostras estava de acordo com os limites legais. Observou-se acréscimo dos níveis de caseinomacropeptídeo (CMP) no leite UHT, provavelmente em função das proteases de bactérias psicrotróficas. Conclui-se que a qualidade do leite cru influencia diretamente os valores de CMP.(AU)

ABSTRACT Studies aiming at improving the activity and stability of dispersed W and Mo containing Pt catalysts for the CO tolerance in proton exchange membrane fuel cell (PEMFC) anodes are revised for the following catalyst systems: (1) a carbon supported PtMo electrocatalyst submitted to heat treatments; (2) Pt and PtMo nanoparticles deposited on carbon-supported molybdenum carbides (Mo2C/C); (3) ternary and quaternary materials formed by PtMoFe/C, PtMoRu/C and PtMoRuFe/C and; (4) Pt nanoparticles supported on tungsten carbide/carbon catalysts and its parallel evaluation with carbon supported PtW catalyst. The heat-treated (600 oC) Pt-Mo/C catalyst showed higher hydrogen oxidation activity in the absence and in the presence of CO and better stability, compared to all other Mo-containing catalysts. PtMoRuFe, PtMoFe, PtMoRu supported on carbon and Pt supported on Mo2C/C exhibited similar CO tolerances but better stability, as compared to as-prepared PtMo supported on carbon. Among the tungsten-based catalysts, tungsten carbide supported Pt catalyst showed reasonable performance and reliable stability in comparison to simple carbon supported PtW catalyst, though an uneven level of catalytic activity towards H2 oxidation in presence of CO is observed for the former as compared to Mo containing catalyst. However, a small dissolution of Mo, Ru, Fe and W from the anodes and their migration toward cathodes during the cell operation is observed. These results indicate that the fuel cell performance and stability has been improved but not yet totally resolved.

ABSTRACT

OBJECTIVES: To evaluate the mechanical properties and metallurgical characteristics of the M3 Rotary and M3 Pro Gold files (United Dental). MATERIALS AND METHODS: One hundred and sixty new M3 Rotary and M3 Pro Gold files (sizes 20/0.04 and 25/0.04) were used. Torque and angle of rotation at failure (n = 20) were measured according to ISO 3630-1. Cyclic fatigue resistance was tested by measuring the number of cycles to failure in an artificial stainless steel canal (60° angle of curvature and a 5-mm radius). The metallurgical characteristics were investigated by differential scanning calorimetry. Data were analyzed using analysis of variance and the Student-Newman-Keuls test. RESULTS: Comparing the same size of the 2 different instruments, cyclic fatigue resistance was significantly higher in the M3 Pro Gold files than in the M3 Rotary files (p < 0.001). No significant difference was observed between the files in the maximum torque load, while a significantly higher angular rotation to fracture was observed for M3 Pro Gold (p < 0.05). In the DSC analysis, the M3 Pro Gold files showed one prominent peak on the heating curve and 2 prominent peaks on the cooling curve. In contrast, the M3 Rotary files showed 1 small peak on the heating curve and 1 small peak on the cooling curve. CONCLUSIONS: The M3 Pro Gold files showed greater flexibility and angular rotation than the M3 Rotary files, without decrement of their torque resistance. The superior flexibility of M3 Pro Gold files can be attributed to their martensite phase.

@#<p style="text-align: justify;"><strong>BACKGROUND:</strong> Human milk is considered the optimal nutritional source for infants. Due to the possibility of microbial contamination during collection and handling, milk is pasteurized to prevent transmission of pathogens. In low-income areas where pasteurization is inaccessible, the pursuit for the best alternative in rendering donor milk safe remains.</p><p style="text-align: justify;"><strong>OBJECTIVE:</strong> We aimed to determine and compare the efficacy of flash heat treatment and holder pasteurization in preserving human milk IgA while reducing bacterial contamination of donor breast milk.</p><p style="text-align: justify;"><strong>METHODOLOGY:</strong> This is an experimental study which utilized pooled donor breastmilk from healthy mothers later subjected to bacterial analysis and immunoglobulin A level determination prior to and post flash heat treatment and pasteurization. Standardized scores were used to normalize population with unknown parameters. T-test comparison of means and Levene's test for equality of variances were used.</p><p style="text-align: justify;"><strong>RESULTS</strong>: Twenty samples of aliquoted breastmilk were subjected to pasteurization and flash heat treatment, both yielding a statistically significant reduction in colony-forming units using Blood agar and MacConkey plates. These sample groups also underwent IgA level determination using Bindarid Kit IgATM and there was no significant decline in IgA levels.</p><p style="text-align: justify;"><strong>CONCLUSIONS:</strong> Flash heat treatment may be an alternative for holder pasteurization in providing safe and effective breastmilk.</p>

Objective@#To evaluate the influence of different heat treatments on the corrosion behavior of a new Pd-Ag-In-Ga alloy. @*Methods @#The corrosion of a new Pd-Ag-In-Ga alloy was tested according to ISO 10271:2001/cor.1:2005(E). Taken as comparison, a commercial high-palladium alloy (Spartan® Plus) was also employed in the study. For electrochemical corrosion test, the open-circuit potential (Eocp) and potentiodynamic polarization curve were recorded and then the electrochemical parameters, namely corrosion potential (Ecorr) and corrosion current density (Icorr) calculated. The surfaces of the specimens that before and after corrosion test were observed by scanning electron microscopy (SEM). @* Results @#Both of the high-palladium alloys had lower corrosion tendency and higher corrosion resistance by heat treatment in the ISO 10271 electrochemical corrosion test. For both alloys, heat treatment at 500 ℃ was found to be effective in improving the corrosion resistance as the best datas of electrochemical parameters (Eocp, Ecorr, Icorr) were detected among the four temperature groups. However, no evidence showed that there was significant difference in corrosion resistance between the two alloys. Comparison of surface morphology of the specimens before and after electrochemical experiments by SEM demonstrated that all groups of the two alloys were visible of pitting corrosion and crevice corrosion phenomenon that gradually became less with increasing heat treatment temperature. Horizontal comparing among the groups of the two alloys, no apparent differences in the distribution of pitting and crevice corrosion was found. @*Conclusin @#The variation in compositions of high-palladium alloys did not cause significant difference in corrosion behavior due to the identical palladium matrix. The similarity in corrosion behavior among the two high-palladium alloys might be attributed to the predominant palladium content.

ABSTRACT

[Objective] To observe the clinical effect ofWen Yang Zhu Shui Formulaecombined with moxa salt packets in the treatment of malignant ascites. [Methods] 61 patients were randomly divided into an observation group(n=31) and a control group(n=30). The patients in the control group were treated with western medicine, while patients in the observation group, based on the treatment of control group, were treated with Wen Yang Zhu Shui Formulaecombined with moxa salt packets for one time a day for two weeks. The effective rates for malignant ascites and Karnofsky performance score(KPS) were compared in the two groups. [Results] The effective rate for malignant ascites in the observation group(80.64%) was higher than that in the control group (53.33%), P<0.05. The observation group showed significantly higher score of KPS than patients of the control group( P<0.05). [Conclusion]Wen Yang Zhu Shui Formulaecombined with moxa salt packets can effectively reduce the malignant ascites.
